People do it all the time. They rarely, though, go straight from spores to cross-breeding. Usually, what you want to do is make a series of dilutions of your spores in sterile water to increase the likelihood of getting monokaryotic growth on agar. Once you have two monokaryotic cultures, put them on the same plate and see if they mate.

Off topic in advanced mycology.

Dikaryotic mycelium will also cross with other dikaryons.  This happens with every multi-spore grow, whether or not somebody named them the same or differently, or if they came from different prints or syringes.

In the book 'The Mushroom Cultivator' by Paul Stamets, on page 27 figure 28 there is a petri dish containing 4 strains of Psilocybe Cubensis mycelium. They are as follows, Matias Romero, Misantla, Amazonian, and Palenque. It's possible. Definitely pick up this book from the library or book store. GREAT read.
