What are your thoughts about Test Tubes?

I found RR's DVD and lipa's stock culture tek helpful. I was lucky enough to find a lot of pyrex at a tag sale a few years ago!
I have about 70 small tubes 12 of the medium 9 of the largest
How would you use these different size tubes?

To find test tubes locally, go to your city's hospital district and find a medical supply house. They'll either have or can order test tubes and petri dishes.
You would treat larger tubes just as smaller ones except for the amount of agar and the size of the piece of wood you put in each. RR

Lab supply places have them, but they tend to be hard to fine locally IME.
aris, they are all screw cap yes? Screw tops are the best. You *can* do it with straight mouth ones and stick a piece of polyfil in covered with foil, buuuuuut one wrong move and that will fall out in not sterile conditions. I test tube bung may provide better results, but you'll need to figure out some way of providing a little gas exchange.
All the sizes should all work the same and be used the same per RR's tek on the video. I use large ones for my culture storage because I found some super cheap on ebay. I like them because they are easy to get culture pieces out of, but the small ones will work just fine provided you have small enough tools... which shouldn't be an issue.

they also sell slip on caps to cover test tubes

search for polypropylene test tube closure or test tube caps

Thanks for the tips everyone!
I wish the test tubes had screw caps but alas, they do not. At least they were almost free!
Whats the best temperature to aim for in the stock refrigerator?
How often do I have to let some fresh air into my stock refrigerator?
Does light influence long term storage of cultures?
Is it ever best to freeze a culture?
When I have a minute I will add insulation to my mini-frig maybe.

for storage you want below 0F but you want the temp to be steady like a deep freezer, a refridgerator/freezer cycles on and off which can destroy the culture. you dont need to provide any air exchange because the culture will be dormant, I've used distilled water and mineral oil for topping my slants distilled water is preferable

Many species will be killed by storage below 0C/32F. Other species such as Shiitake and oyster can be readily frozen. One needs to be sure first. I keep my culture refrigerator at 1C/34F. The slants are all in insulated lunch boxes to ameliorate the temperature swings. RR

I saw another user post this link when they were talking about how they make their self-sealing lids (they used the blue rubber stoppers). They also have plastic tubes, and those test tube lids that were mentioned. Even if privacy were an issue, I think I would prefer to buy from a place like this, and maybe have it shipped to a middle man. Hope that helps.

Hmmm... I have had quite a few cultures destroyed by the fluctuation of my mini fridge. The insulated lunch boxes are a great solution! RR, what sort of lunch boxes are the ones you use? Are they the soft ones?

What is the advantage of storage on agar, instead of suspension in distilled water? * An original goal here was to fit more samples in a smaller amount of refrigerated space. I have no idea how fast a para filmed test tube, full of h20 (rather than agar) evaporates at 34F (or room temp for that matter).

I've been wanting to jump on the cyrovial bandwagon myself... They are pretty close in price to glass tubes, and can be autoclaved just like'em... Inside threads... DNA and Genetic info free...
Sounds like the perfect storage...
And 5 years is a LONG time lipa!
