LC is a nutrient solution in which mycelium can grow.
mycelium suspended in water won't be a LC, it'd just be myc in distilled water.

injecting some water in a grain jar won't leach that much nutes, but if you're concerned, after you save your myc suspension let it settle, then suck out water and add more distilled water. repeat if necessary.

it is either me not getting your issue right or you not understanding that LCs cannot be stored for longer than mycelium in distilled water.

if you want to store it for years, you want it to be hydrated but without any nutrient. as LC stands for Liquid Culture, it means it contains nutrients and is a culture of spreading mycelium. and perpetually culturing mycelium doesn't seem to me as better than having it in suspended animation in pure water.

vials are preferred due to their smaller size but if you want to keep a gallon of mycelium suspended in a jar of water go ahead  it's just overkill though as if storage goes wrong you lose all at once, while a few mls divided through separate vials ensure higher chances of viability.

It all depends on species.  Some, such as oyster mushrooms can be frozen solid and last indefinitely.  Others, such as Shiitake will live for a decade or longer under refrigeration if you have wood in your master slants.  Once mycelium colonizes wood, it can remain dormant and survive for a very long time.

you use the word semantics a lot. I guess you like it.

Does creating the slurry from a grain jar using distilled water produce a non-nutritious medium, or will the distilled water no longer be distilled, but instead now contain nutrients after it touches the colinized grain? (albeit for 30 seconds as you swish the grain/water around)

anyway, my answer to your question is: the water you'll draw out of grains will contain some nutrients so it will be less effective than distilled water alone at storing your mycelium.

also RR raised a good point on using wood.

Could you use a Q-tip or cotton swab to fish out some mycelium from the slurry/grain, and deposited that mycelium in the distilled-water-long-term-storage vessel of your choice?
How much nutrient solution would hitch a ride in that case?

I too am interested in how people have reached a totally non-nutritive distilled water solution, if using mycelium off colonized grains.

One just needs to cut out a piece of an agar wedge, and store it in sterilized distilled water...
This will allow atleast 5 years im sure... :super:

So, do cultures stored in dH2O need a gas exchange port? Still wondering how to create my jars if that is going to be my storage vessel. I do have vials with caps, but a larger amount of LC would make me feel more comfortable.

In the paper i linked up above, they stored their cultures in cyrotubes that had teflon washers, making an airtight seal and PREVENTING evaporation...

I really would like RR to chime in here at this point, because i know i have seen him say multiple times that you need air exchange when storing cultures...

PS... Sorry for talking to myself in this thread!

Apparently the cultures go into stasis.  I have no direct experience other than I have a culture syringe of Pan stipticus that's 8 years old and still very viable.  I use a drop or two every few years to start a petri dish, and then put it back in the fridge.  So far, it's still good.
