Rye Grain Bag Colonization Time * 1
    #14594984 - 06/11/11 08:54 AM (12 years, 7 months ago)

How long does it take for first signs of colonization in a 1lb rye grain bag? I inoculated it on 6/4 and still have no signs of colonization. Also how much FAE do I need to give it?

Don't give it any unless you want contams and you could have another week before you see any growth.

does your rye bags have a filter patch? how were you planning on giving it fresh air anyway.I think you need to search and read A LOT more

Did you shoot it with a spore solution?

This will add a week or so before visible growth (maybe more
maybe less, but more time anyway)

Your impatience is natural.  Fight it!

I thought air flows through the filter patch. Anyway, yes I need to fight off my impatience. I have been reading threads and I think I have found my error. I injected the solution directly in the middle of the bag. Should I wait until day 10 and if still no signs of growth, re inoculate?

It can take up to 3 weeks on the outside for MS,your still in the early stage yet and the filter patch is for GE not FAE

The filter patch on the bag allows for gasses to exchange.  Gas exchange is required for colonization while full exchanges of fresh air are required during fruiting.  The pore size of the filter patches are too small to allow for full exchanges of fresh air.

Since you innoced. down the center of the bag, you may have germination now but aren't going to be able to see it until the myce. grows out and into the sides of the bag.  That's why it's prescribed to innoc. down the sides of the container so that the first signs of myce. germination can be observed and monitored.

Don't shake it if you inoc with MS,mixing the bags after inoculation is for GLC,LC

MATTER OF PERSONAL PREFERENCE.!

More of an advanced method but, it's as ok to "shake" upon innoc. when innocing. with MS.  Distributes moisture evenly and spreads them spores around.  Spores are strong enough to survive the vacuum lock of space.  A little "shake" (gentle roll on a table) with a freshly innoced. MS sterile grain medium isn't going to disrupt a mushroom spore.

As far as spreading the spores to far apart to make mates, POPPYCOCK!  Spores are microscopic.  There are billions upon billions of spores in any given syringe, even if their not visible to the naked eye.  Mates WILL be made.

Mates will be made but it will take longer and then there will be a rash of "why are my spores taking so long to germinate"threads also its nice to give a squirt down the sides so you can see the first germinations

I thought that the consensus was clear on this.

MS inoculations require germinating spores to find each
other.  This makes mixing right after inoculation a bad idea.

Live culture inoculations can be mixed immediately.
(watching a grain sack explode when using an LC is great!)

Take care,

JD

This is incorrect.  Don't spread false information just because you have no experience with any particular method.

FACT; colonization DOESN'T take longer when shaking upon inoculation.  Germination takes the same amount of time, it's just germination comes from the middle of the medium.  The only thing that takes longer is, MYCE. GROWTH BEING ABLE TO BE OBSERVED.

If a "rash" (to use your words) of questions happens because people shook MS grain mediums upon innoc., they either shook to hard, syringe was contaminated, or poor sterile procedure.  Can't go blaming the methods that have been proven to work because someone goes and messes up the procedure.

I don't feel people are stupid and I choose to give em' the benefit of the doubt when I choose to present them with advanced methodologies, until they prove me otherwise.  Mycology isn't rocket science.  It's just something new that needs to be learned first.

EDIT TO ADD= P.S. JD, I DON'T follow the crowd.

So should I re-inoculate after 10 days and no growth?
Also I have extra pH neutral soil from my grow, would I be able to spawn to this? Its sunshine Mix #4 and I mixed in some earthworm castings, High N guano, and some bloom fertilizers. All organic though. Please reply to this, no arguing please.

No you can't spawn cubes to soil, they need easily digestible amino acids, sugars, micro nutrients that just aren't found in soil alone. You also can't use raw fertilizers like Bloom, mushrooms don't use fertilizers like plants do
