Spore Clump in Syringe

Hey guys, I've read through some past posts but have only found posts related to multiple spore clumps in a syringe. I have 8 jars I'm about to inoculate, but my syringe has one fairly large black spore clump in it. I've tried to shake it but it doesn't seem to break up. So is it OK to inoculate after shaking the syringe even if the spore clump hasn't broken up? Or does it need to break up? Thanks guys. If I've missed a post where this has been answered, I apologize.

try pulling the plunger back to let in a little air, then shake the shit out of it!
if for some reason it will NOT break apart, you'll be just fine.
grats on getting a syringe with what sounds like a ton of spores

Be sure to heat the syringe until its red hot before you suck in air if not you will not be sterile any more and could introduce contaims .

Do not do this unless you want a side of contamination with that little bit of air.
"Shaking the shit" out of the syringe is fine before inoculating, even if you have a clump of spores that won't break up.
Spores are microscopic.
I can assure you that if you were to observe the solution under a micro-scope, there would be billions upon billions of highly active spores through out the solution that can't be observed by the naked, human eye.

i get my syringes from the hawkseye and everyone has massive clumps in them and they have worked just fine the hawkseye is the shit fast shipping good viable spores. although its really not necessary they do it just to please their customers because most people think its better and i guess people were paying more for dark spores syringes at other sites so now its just their standard to make all their syringes extra dark

i've never had a contam as a result of doing this. we've never had contam PERIOD...so yah
obviously don't pull your plunger back in a dirty environment. we do all the work in a pretty badass glovebox
we also do G2G in the same glovebox and haven't had any contams O.o

Hey guys, thanks for the replies. Yeah, I knew I could suck in a little air with the tip of the needle red-hot, but I'd still be worried about contams. If I was to do this, would I be sucking in air while the tip of the needle was red-hot and in the flame? Or do I take it out of the flame and while it is red-hot take air in? And also, would there be a problem if I used my gas stove flame, since it would be harder to do while lighting a lighter in one hand. Thanks guy, great tips like always.

My Hawks Eye syringe also had the clump. I failed to shake it and had 10 out of 40 innoc holes not germinate. My next syringe I shook and beat against my hand until spores broke apart darkening the water.

I know this is old but if you ever run into the problem again, just tap the syringes against a some surface (the inoculation glove box is perfect) and they will all disperse evenly. It works better than even shaking very vigorously, you can practically slam them and they don't bust or break.

Definitely don't do this. I followed this advice tonight and after the second tap I felt myself getting showered in a mist of spore solution. One spore syringe down the tubes... Cracked right down the full length.
Hit it against your hand. Don't risk smashing it.

The only time I have trouble breaking up the clumps is if the syringes had been pulled right out of the fridge before inoculating. Allowing them to come up to room temp does the trick for me. That and shaking them like a red headed step child BEFORE putting them in your SAB (limited shaking mobility). Clumps or no, you are good to go after shaking. Do not draw air in, even after sterilizing the needle. Not a good idea, even in front of a flow hood.

I just shake, whip lid or what ever, flame sterilize, then I knock up those plates or jars.
11 Jars and 20 plates off 1 syringe and with my first time doing agar im hoping atleast one of my homemade pasty plates will work.

What works for me is grabbing the syringe like a testube and tapping it with a finger. You hold the syringe between your index and middle finger and your thumb, hold it sturdy and then start flicking the bottom of the syringe with your index finger from the other hand. Usually this breaks up the clumps but if it doesn't I just shake vigorously in every direction until most of the clumps have dispersed
For the record, all spore syringes are dirty in some way or another. Its good to take sanitary precautions but don't go around with the idea that syringes are clean
