Culture Slants- To tighten the lids or not?

Hey all,

I know it's probably been covered a hundred times, but I can't seem
to find the threads...

I've got some culture slants stored at room temp as backups for the ones stored in the fridge.
They are about 2 months old and have lightly loosened caps as well a cling wrap wrapped around the lid.
Some cultures are so vigorous that they are breaking through the cling wrap barrier and growing through the ziplock baggie.

Any suggestions on whether I should have screwed the lid on tight after the tubes colonised???
I was under the impression that a lack of air would have killed or compromised the culture.

Also if you have a better way to store cultures at room temp. I'm keen to hear all recommendations!

If you keep the culture at room temperature, of course it's going to keep growing. Are you just keeping these as insurance in case your refrigerator goes out? If your fridge goes out, then cultures inside become just like the ones that you're keeping at room temp and you've neither gained nor lost anything. A better way to keep you redundant copies is to give the backup slants to a friend who can keep them at 2-4C.

I've heard of people having success by storing mycelia in sterile water, but I don't recall if it was refrigerated or stored at room temperature. Even so, sterile water suspensions kept at room temperature can only be more successful than slants because agar slants at room temperature have zero chance of success over the long term.

I've had good luck with distilled water storage.  They're crucial for heat loving species that will die in a fridge.  I just recently tested one culture that had been in storage about a year at room temp and it woke up fine.  Probably varies by species.

ah great work.
I've seen a few different teks, what one did you use,
sounds like it worked a treat!

I've read that it's essential to fill the test tubes to the brim,
make sure they are totally devoid of air.
Is that true?
What worked for you?

I did nothing so complicated.  Steps I took were something like:

1) Fill tubes with roughly 50% of water with distilled water
2) PC for 30 minutes or so
3) Inside still air box use sterile scalpel to 'shave' mycelium off plate and put in tube

I even had the cap on tight.  Not quite sure why it didn't die since the agar slants I did with the caps on tight totally stalled out.  Maybe because they're immersed in water?  Not sure really.

From what I understand I think that the water storage tek
slows myc growth down to a crawl.
It can survive with minimal oxygen in that state for years.

I'm going to try the water storage tek today,
see how it goes!
