keeping cultures for long periods
    #14519616 - 05/27/11 07:27 AM (12 years, 8 months ago)

(Everything done sterile)
I removed the tissue and I let it alone for a few days to see if the myc would grow from it. 1 of 3 samples started growing white mycelium. Yesterday I pressure cooked the 4% honey and water mixture, and today I transfered the sample to the mixture. Ive left it in the dark near a radiator.
Assuming this suceeds, how would I get the most grows out of this? I have 100ml of it. I could always make some new jars and inoculate them from the master jar... But the point is how could I keep that master jar for lets say one year? Is this possible? Because if I keep making cultures from the previously made jar, eventually the isolate will weaken (if I understand correctly) unless I change the medium every time I transfer...
Ideally, every time I decide to grow, I take 1 ml of master LC and inoculate another 20 ml or so to then inoculate rye jars. this way the master jar would last 100 grows (if there is 100 ml iof master LC)
Any ideas please!

Room temp, not by the radiator.

If it colonizes correctly and tests well, refrigerate most of it, use that as a master culture to start new cultures.
But test it very well first.
