If your talking about PF tek jars then do four holes evenly spaced for four inoculation points.. You COULD try one but its not going to colonize very fast, plus other stuff.

If your using a spore syringe then 4 holes are better for more even distibution of inoculant,if your using LC then one inoculant hole and shake. You still need a larger 3/8 hole covered in tyvek, poly, micropore tape for GE

What other stuff? I inoculated my PF jars using one hole in the center. Probably should have done 4 because of the GE deal, but can punch three more holes in the lid to make a triangle w/ one hole in the center. Has anyone ever tried just using one inoculation/GE hole for the PF Tek and get good results? Thought I'd post on here instead of starting a new post.

You can and still get good results depending on how your MS goes, but if were talking optimal then its four holes for inoculation and one in the centre 3/8 to 1/2 inch

If you're using WBS, all you need is a single 1/4" hole in the middle. Stuff it with polyfill. Cover that with good quality Tyvek then put the ring on the jar. With WBS, you shake to distribute the spore solution or liquid culture, so you don't need multiple holes.
