Spore solution to Agar

Does it matter if I put spore solution on top of the agar without penetrating the Agar? Also how long does it take before signs of growth show on agar when using a spore solution?

you can use spore  or tissue.
just get one drop in the center and you will be good.
it can take 3-7 days sometimes longer.

less is more when using spores on agar.

If anyone is interested I put 4 drops of spore solution in this agar (which is specially made for fungi) without penetrating it and these are my results after 2 days
First one

I got sloppy with the second one

look at the three slits on the lid , its called a vent,
you see the space between the lid and the bottom around where they meet, air can get there,
go look how to wrap a dish properly on youtube or something youtube has everything, its scary...
untell then leave the agar alone, so you dont waste any, and a tip , when pouring agar in dishes use a syringe to squirt agar in

that looks like bacteria or something you don't want. It's not mycelium.

These don't have slits. The lab I work in deals with enzymatic disaggregation so we have special ones. As for that being mycelium or not I'm not sure. Also, I did use a syringe (I'm just a poor shot) how else would I have done it?

iam not sure about your dishes. it could be bac.. give it a few more days and see what u get. did you make ur or is it from a vender?
this is a 2nd transfer i did working on an iso.

thanks guys, The spore prints are from wild cubensis. I don't think it is bacteria because it's growing exactly where the solution hit the agar.Then again the spore solution may be contaminated. I guess ill find out sooner or later.

your going to get far in this hobby lol dont like to a admit when your wrong, when the ppl that are telling you are the ppl who have been doing this forever.

well someone must have figured out how to do it because every cube use to be wild right?
Are you saying that every time I take a spore print from a wild cube and inoculate agar or substrate they will become contaminated or there is just a high probability it will?

try taking a tissue tample.
from the inside of the stem that has never been exposed to air. of course.

I don't believe I ever said I wasn't wrong? I openly admitted I didn't know if it was mycelium or not which is why I posted pics. Also, I'll make sure not to use electrical tape next time.
