spores and strain evaluation

Trying to find out some answers to a few different questions regarding strain evaluation as well as creating new strains.

If one was to take a spore print off of a cold weather variety of say a shiitake, would the resulting multi spore germination, and sectoring to create a new shiitake strain create another cold weather strain, or would it be up to chance on the temperature preference?

Also does the spore print that is taken from a strain that is old and worn out and showing signs of senecense also compromised or are the germinated spores re-invigorated and fresh?

Cultures grown from spores should be totally fresh genetics.  Senescense comes from the mushroom tissue expanding until it gets old and runs out of energy.  Spores are a new generation.

Growing out spores from a specimen will probably tend to share some of the characteristics of the parent.  But of course it's not that simple.  If you have a cold weather strain and take a spore print and end up with 10 new sectors it's certainly possible none of them will share the particular temperature trait as the parent.  You would have to grow each one out and judge them on their own merits.  And if they all suck start over.  This complication is why most folks end up buying trusted cultures where someone else has done all that busy work.

In addition, it's really hard to get a great fruiting strain from spores.  You can isolate 100 cultures, and there's a good chance that all of them will be non-commercial quality strains.  This is why most growers select a strain which matches their conditions.

I'm not trying to prevent experimentation, but be aware of what you're getting into.  It may take a long time, and you never know how any isolate will perform until you've tried various colonization temps, cold shocking techniques, fruiting temps and humidity, substrates, etc.

In principle, the genetic variation among the offspring depends on how different the parents of your shiitake were. If your strain is the result of a long line of inbreeding and selection, you will get a lot of strains with almost identical features. If it is the result of crossing two strains with numerous genetic differences, you will see many different strains.

With oysters I get great results by inoculating a few small substrate jars with spores, fruiting them and just cloning the fruits I like. This is not as controlled as the usual method and I have to repeat fruiting and cloning as the first clones are rarely only one strain, but I still end up with fast colonizers and excellent fruiters.

With shiitake it is said to be harder, so - as RR wrote - you may have to fruit dozens of small pf style cakes.

hmmm, great answers everybody!
I am interested in new strains for a few different reasons.
One, should any disaster destroy my decent strains I will have backup spore prints to start new strains (it's much harder to get ahold of decent strains here in Australia)
Also just for experimental fun!

I was quite motivated when I read about RR's shiitake strain that was an extremely cold temp. fruiter!

I think the more we germinate spores and experiment, the more selection of strains we'll have to match up with our climatic conditions, the more resilient mushroom cultivation will be! Then we can simply swap strains of mushroom species to suit the season!
