I had a big fat weird fruit, so I thought I'd try my hand at cloning...
...took 4 samples...  this is one of them...  is it an isolate??
I can see a slight variation though, in the 12-3 0'clock area...  could this possibly be another sub-strain?


I think I'll try a transfer from this sector tomorrow...

That huge, weak gap from 3-4pm doesn't do it for me. I'd transfer a few more times.

Ideally, you'll have equal growth all the way around in a good isolate.

Its hard to tell from the pic, but if that is a clone with no transfers, and you
started with MS then that cannot be an isolate.
I just did my first transfer, and the piece of mycelium that I transfered from was less than 7/16" in diameter.
Make your first two transfers when the mycelium is really small and young before
it has a chance to exchange dna with another strain.
Then a couple more should get you to an isolate :smile:

taking tissue samples from fruits growin from multispore can often contain mycelium from multiple ginetic sources as in multispore grows fruits can be made up from several different mycelium networks.

So just transfering to agar wont produce an isolate.

Looks like there is 3 sectors there. So you will have to do some transfers. its probably best to take from the strongest looking sector which is the top section from 9oclock til 3 o clock.

Just for the record, I think it's an average of 4-5 transfers that will net you an isolate. You'll know it when you see it, because it'll grow at the same speed on all sides and you won't see distinct sectors.

I'd take 3-4 samples from that dish, 9-3pm positions, transfer, and see what happens. Agar is fun. :biggrin:

No, you should take samples from the 9-3 area to make 3-4 new petri dishes.
This sector seems to be more dense or healthy.

Is there a little yellow spot between 7-8 o'clock on the edge of the agar?
