I have food club light corn syrup with real vanilla. I plan on buying rtv silicone and drilling a hole in the lid and then just using that to seal the hole. Then i will inoc them in a still air and let them cultivate in there.

bear in mind if you just have 1 hole that is sealed with silicione, once you pressure cook it, its going to have a vacum once cooled down.
If you dont equalise the pressure, as soon as you stick the syringe in, its gonna suck the contents of your sysringe into the jar.

Well, you could stuff the hole with polyfil (very tightly), cover it with tyvek, or get some synthetic filter discs.
For my LCs I do two holes covered with silicone, then I just use a syringe minus the plunger stuffed with polyfil in one hole. Aspirate out of the other.

You could always PC your jars with a syringe in the inoc hole without the plunger. It would equalize pressure on its own once its cooled.

If you have two holes one with rtv one with poly, innoc in the rtv port so it heals itself. As for the poly hole it should equilize the pressure in the jar.
You could def do two holes like you said and just cover one of the holes with micropore tape and that should do it as well.
