mycelium thinner on agar
    #14442681 - 05/12/11 10:27 PM (12 years, 9 months ago)

So, I've moved some mycelium tissue from a brf cake to a petri dish in an attempt to isolate a strain.

After moving the tissue, which was very rizomorphic, and it continued to grow out for about a millimeter. After that, it got much thinner, but continued to grow. I'm not sure if what I'm seeing is a weaker strain that has overtaken the thicker mycelium, or if it's just a lot thinner because it's on agar and there aren't as many nutrients available.

Does anyone know which is more likely? I've transferred 5 different pieces from different cakes, and they're all acting the same way.

You want to start from spores on agar to isolate vigorous mycelium strains with different characteristics.

By the time the mycelium was taken from the BRF cake it had already lost some of it's potential (not much, but some). It had also become pretty much a singular entity composed of poor and great fruiting strains, becoming average.

Start spores on agar and begin taking samples to transfer pretty quickly after the growth starts. Do this several times and you should begin to see clear sectors representing different strains.

Changing your mind is one of the best ways of finding out whether or not you still have one.

Check out my post in the cultivation forum and check out the rye seed jars I inoculated with the petri dish. It's growing very rapidly. I have two strains isolated. =)
