my under $20 agar kit

whats up shroomery....

i was inspired by a member here, who i am almost certain is not actually licensed to practice medicine, to get some "grocery store agar" action going on..

so i spent 20 dollars and 30 minutes getting what i needed to get started.
i already had a SAB, cutting and sterilization stuff, etc.

agar ws a whopping 8.92 for five packs at the local asian supermarket where i get the hookup b/c i go there alot for work....plus the cashier's are super hot :smile:

i spent another 10.98 at a big box store on two packs on instant potatoes, four half pint wide mouth mason jars, and a 5 dollar tkae and bake pizza. i always get mad baked before going mycology shopping so i figure munchies really are part of the kit expense.

attached are some pics...wow that was alot easier than i thouht it would be. now to clone the shit out of some of the first matured shrooms from my first flush!!

Get something like this 500 Polystyrene 100x15mm ~70$ or 24 Pyrex 10x15mm bottoms ~37$ and 24 Pyrex 10x15mm Covers ~40$ you will be 100 miles ahead. Light malt from your local DIY brewing store.

cool man, thannks. i checked that ebay link and that seems like a hell of a deal. figure i can start with the odd wide and short half pints for now, but i did order a 24pk of those glass petris.

for now the plan is to stick close to the grocery store agar tek and clone off of the first mature fruit bodies from the PES amazanoian i got in some MSH tubs now. I figure i can fuck around with the clone on agar until i got some kind of cool super aggressive myc. isolated over a couple weeks.

my goal with the agar is to fill a mini fridge i have with badass cultures on agar...


in the meantime i need to figure my agar recipe...been doing some reading and finding some teks but trying to keep it as simple as possible.

also need to be sure of PC time on the agar in jars...i am going to mix it and then PC it in the jars to avoid possible contams from the pouring procedure. i dont see how i could do the pouring in my SAB and i dont have my flow hood finished yet so i am thinking this will be the safest way to get it done...

2 very simple agar recipes, that are easy to source the ingredients for are,

MEA (Malt Extract Agar)
10 grams LME.
9 grams powdered agar agar.
500 ml water.

PDA (Potato Dextrose Agar)
5 grams dried potato flakes.
10 grams powdered dextrose.
10 grams powdered agar agar. 
500 ml water.

PC agar mix in jars for the same amount of time you would if you were PC'ing agar mix in a large container to poured into petri dishes.  I use 35-40 minutes at 15 psi for the sterilization all my agar mediums.

pouring in your box is kind of tricky until you get the hang of it. i normally put my PC next to my box and let the MEA mix cool enough set it in the box w/o leaving condensation on the lid of the box. then i leave it sit in the box after being wiped off w/ 91% alc. and spay air sanitizer. It sits with the 20 plates for about an hour or 2 depending how hot the MEA is yet. the key is to let it cool enough that when you pour the plates there will be little to no condensation on the lids. Always wear gloves and long sleeves and cuff and tuck the wrist of your long sleeves into your gloves as skin cells will contam a plate because humans are contaminated disgusting creatures like that. when ur ready and after ur cleaning and prep work stack  your plates in workable groups so that you can pick up the stack of plates with one hand and pour with the other. pick up the bottom most cover for the bottom most dish while picking up every plate above it pour that bottom dish and place its cover on then pick up the next cover above it leaving its bottom resting on the cover of the plate u just poured. repeat until you have a stack or 3 of poured dishes all resting ontop of each other. after they cool 3 hours they should be ready for work or you can wait 3-4 days to ensure there are no contam on the plates.

(I think) with the plates u have (masonjars) you can put the lids on and just PC them as is with the agar mix (1/20th dose of 10mg agar 10mg LM and 500ml) all in the jars. let them cool in the PC overnight and they should come out fine but i have never worked with these jars so i cant say for sure.

top shelf info here guys :cheers: Also, I must remember to incorporate munchies into my kit budgets :P

A question, if I made syringes, inoculated 60 jars etc without contams in my GB, should I be able to pull off agar with the same procedure?

:yesnod: but make sure to wipe down your agar jars (if using the 1/4 pint ball jars) before putting them in your GB. Also, if pouring the jars instead of just pcing the agar mixture in the jars & letting them solidify like that, make sure to wipe down the bottle before putting that in your GB too.

Those packets are 0.88 G correct? I'd try your local health food store to see if they have it there. I got 2 oz. (56 G) for $7. 5 packs @ .88 G = 4.4 G. for $8.92. I thought $56/lb was expensive (health food store). You're paying $2.02/G. $905/lb.

Somebody check my math,please.

thanks all!

im gonna get through the work stuff this weekend and then bust out the agar on monday.

will keep ya'll updated on the agar experimentation.

also stopped in a livestock supply place and got a 60cc luer lock syringe and three 16ga 1,5" needles for less than $3!!!

Yes.  One thing to keep in mind is that you won't have as much vertical height inside of a still-air-box as you would in front of a flowhood.  Keeping this in mind, be sure to check the size of your agar container by "air pouring" the agar (pretend your pouring agar into petri dishes) inside your air-box to be sure you can comfortably pour agar into petri dishes inside of your still-air-box. 

Back when I was confined to pouring agar in a still-air-box, I used a regular mouthed, pint-sized jar for the agar vessel.  Smaller jars can be used.  I'v read of others using a short and stout whiskey bottles, such as empty Crown Royal bottles, be creative.

Height is one problem, condensation is the other. It's a big part of why I went to using jars.

really, height in the SAB/GB?...come on guys, do a bit of searching. I can stack 2 quart WM jars on top of one another in mine...height is in NO WAY an issue, if you have a proper SAB/GB...71 qt :woot:
