You do not have to use a glovebox to inoculate jars, but it can help reduce contamination rates if you are having problems with it.

OP is making syringes. You should ALWAYS inoculate BRF jars in a glovebox or still air box. grain jars w/ self-healing injection ports do not need to be inoculated in a GB or SAB.
OP, your house isn't the problem if you're doing it in a properly constructed SAB/GB. Your sterile tek is the problem or you got really dirty prints. transfer them to agar & try to isolate away from the mold.

Well no, I guess you don't have to do everything in your glovebox. But why wouldn't you?
I do my syringes, agar, grain to grain, inoculations, prints, etc in my glovebox.
I would reevaluate your box, if you already have contam in your apartment then you're gonna need to use it. If you hate the way your box is set up now redesign it. I'm on my 4th box now, each one better than the last.

I agree with you on using a still air box or flowhood for any spore work.

make some self healing injection port lids.
you could inocculate next to a moldy loaf of bread. nah, don't do that. but injection ports would allow more successful open air innocculation

A description of your sterile procedure will help greatly, as well.

agar is the only way. NO print is 100% sterile...

yup, that's where it starts. You're going to need to clean your entire house out. Wipe the walls w/ a water+bleach solution & all the riga-ma-roll...
Next time, PC the jar @ 15PSI for 60 minutes before opening it.
I use dishwashing gloves in my SAB/GB

bathrooms & kitchens are the most contam ridden place of your house. My clean room has carpet & I don't have any problems.
so you, put your cap fully submerged into water? That's probably where all the contams came from. That mushroom had tons of them on top of the cap & probably started going bad in the water too...

Take a bottle of unopened water (it's sterile already). Put a dab of silicone on the side (about 2" up from the bottom) for a self healing inoculation port. Cut off a cap that will sit on the rim of the bottle & not fall in. Still don't open the bottle.
Once the silicone is dry, Prep your SAB/GB & put in the cap & wipe down the bottle w/ some iso. Seal the SAB/GB & then remove the bottle cap & place the mushi cap on the open bottle. Leave for 12-24 hours. The mushroom cap will eject the spores into the water bottle.
Come back, take off cap & eat cap. Put cap back on water bottle. Now you've got 16.9 oz of MS spore solution
That's all I can really think of...
or you can me your addy & I'll send you a free PE6 print. I got a few just hanging out @ my house that I'm not going to be using...Might as well share the wealth, so to speak.
