I'd wonder whether a colonised/properly-cased monotub actually needs filtered air - there shouldn't be enough uncolonised nutrients for contaminant moulds to get a foothold (speaking with complete lack of experience).

And if you made a PF-cake and chucked some polyfill in the top instead of tyvek/tape/filter+lid , I'd put money on them turning funky colours with reasonable regularity.  It might filter some particulate out, but fungal spores are too small (in my uneducated opinion) to be reliably stopped by any sort of loose fibre stuffing.

Feel free to check out all the times that people use poly to filter their jar lids. Works great for grains, LC, pretty much everything!

As for established Mono's, no, you don't need the poly, nor do you need anything in the holes as the myc is fairly contam resistant on it's own.. Thats why you can have cakes in a sgfc without contaminating. When you first setup your monotub, you need either poly, micropore tape, or any other type of tape to keep contams out during colonization.

The OP is talking about a partially colonized substrate... Yes, he should have washed it off and then fruited... but he apparently didn't, so the substrate will keep colonizing as long as you keep contams out.

On the first part, yes you are correct. As RR says, "there is nothing sterile about a fruiting chamber". What fruiting mycelia need is not sterility, but fresh air exchange, and lots of it. As for the second, unfortunately not so. To quote RR again, on sterilizing agar:

Put the foil dirrectly on the rack, cake on the foil, then flip the bottle upside down and place over the cakes. That way all you need to do to air it out is lift the bottle up and set aside for a few mins.

If your GH is dialed in, and your cool mist running, all you should need to do is leave the bottle off the cakes for a few minutes to mist and air out. After the cake has been fanned (aired) replace the bottle back over the top and you're golden brotha!
