Agar question (I've already looked around)
    #14393025 - 05/03/11 06:00 AM (12 years, 9 months ago)

So I know the usual response is read more but I can't seem to find this info.
I'm trying to make agar for culturing, but don't want to buy lme or make the potato dextrose.  I suppose I might just do it anyway, but we'll see I guess.

Does anyone know an agar composition that is given in the various vitamins and salts required?  Ive got glucose and every vitamin/salt necessary, I just don't know how much fungi need of each.  I usually do bacterial plates, so my media is all made for that.

Thanks.

P.s. It's not easier for me to just make the potato dextrose:  I already have everything I need in my lab and can make my plates during work.

Glucose is dextrose, for our purposes. Do you have instant potato flakes?

I know glucose is dextrose.  Probably should have said that.  I don't have potato flakes, I guess I could get them, but I'm curious about the actual salts and vitamins required in the media.

Do you have a link to a potato flake recipe though?  I can't seem to find the info I'm looking for anywhere.

Doesn't work like that with the nutrients, fungi expect to break down food and digest it.
You're basically talking about a hydroponic grow, search on that for more info about why it won't work.

Even just for growing out on agar?  Interesting... Also kind of disappointing.  Anyway thanks for the help doc

Agar + glucose would maybe grow something. But the myc wants a starch too.

But I haven't tried every possible combination.

I just decide to use potato + glucose + agar.  Innoc'd with spores and about a week later I have white fuzzy stuff all over.  Now, ita covering the entire surface, but I used a spreader so it should grow that way.  But is this undesirable?  Should I avoid spreading?

It should be covering the whole thing eventually.. the myc growing is good. just watch for contams if they pop up and transfer to whatever your next step is for whatever it is your doing . If your worried about contams popping up in your agar plate just have another one ready you can transfer a little piece of the myc to to try and isolate the myc

There's condensation so I can't get a good pic, but for the first few days, no growth.  Then I look and the whole thing is white.  I manually spread the spore solution and used a bit too much, so this might be why.

I'm doubtful of contams as the other plates are fine and I did it under a biohood with disposable instruments.  I'll open under the hood on Friday and provide a pic.  The possible myc is slightly wet looking, but without a pic I'm sure no one can give me any info.

It's easy to use too many spores in an agar dish.
