Petri dishes
    #14363035 - 04/27/11 07:09 PM (12 years, 9 months ago)

I poured 20 petri dishes a few months ago. I had 4 that I hadn't used. went to use them today and one had mold on it. So I got into the fridge to see if anything else I poured had mold and one did.

So 2 out of 20 petri dishes had mold

I worked in front of a flowhood and would expect the contamination rate to be close to 0. These were the first dishes I have done, and wanted to get some feed back before pour more.

I put the dishes in 2 stacks and poured them. I placed a cup of hot water on each one, and let them cool in front of the flow hood.

My thought is a few spores got through my flowhood while they were cooling, but I'm not sure.

Any feedback is appreciated
Chad

I did have condensation on the lid. This was caused by me pouring the agar while it was still hot. It could be my pouring but I did it in front of a flowhood. When I was cooling them they were not wrapped in parafilm or anything. Just lid on in front of flowhood. Is there a better way to cool them?

You answered your own question in the first sentence, "I poured 20 petri dishes a few months ago."

After that long, anything can get in.  The parafilm will stretch and/or crack, and I've found that after a few months, molds often appear for no good reason.  Just toss it and start with fresh dishes.
