from agar to lc, will it hold the genetics?

so i want to clone one of my fruits coming up, i have all the stuff to do the agar. I was wondering if i get a nice isolate from a nice sized fruit in the center of a cluster and grow it out on agar, could i then put a slice of agar in a lc? I mean will the genetics still be extremely similar once it is introduced into a lc? I dont really have a flow hood, only a glove box and i dont want to add a slice of agar to every grain jar. I think it would be much easier to make the LC with the agar if it works. thanks

why not just skip the agar altogether and do a needle biopsy clone straight to lc ?

ahhh.... very true. damn good call, I even have a couple sterile syringes laying around. Should i run the needle through more than one time to increase the colonization rate?

you could yeah ... just remember each stick is another chance of catching something on your needle you dont want in there though ... as long as you use a glovebox and are diligent in your sterile techniques youll be ok though ... it helps to have a nice thick mushroom for this tek as some of the skinnier hollow ones are a pain in the ass to get a decent plug of flesh ...

don't do multiple biopsies on the same lc...make multiple lc's from multiple biopsies.
The failure rate of tissue->LC is relatively high. Better to make a few. Some will turn cloudy on you. No one is perfect and bacteria are very tiny critters.

ok, that makes more sense. its a lot better to make multiple lc's with only one biopsy shot per lc to help reduce the chance of contam and increase the chance that the lc will succeed

If you really want to up the chances you could go agar->grain ->grain. It only takes a little bit longer to get rolling. But it's a sure bet. Or do both to learn both procedures and see which works for you.

I've never been able to actually pull anything out with a needle biopsy and have now resorted to cutting a chunk out of the base of the stem with an exacto knife.
