air exchange during colonizing

does there have to be air exchange during colonizing? I've been reading mixed reviews. the reason I ask is...couldn't you do the pf tek  without any jars? If done in an EXTREMELY clean environment you could pour the sterilized substrate (bird seed,) into a big zip lock bag and do with that, exactly what you would with the jars? I want to colonize some bird seed but was thinking of pouring the sterilized bird seed from jars into a big zip lock bag and then throw the content of a fully colonized brf jar inside to make a huge batch of colonized bird seed. Figured one big bag would take up less pace then the jars.

It sounds like you're just doing an improved version of what we do anyway for bulk subs.  Thats why we go from grain jars to grain bags to more grain bags or a sub.  It would work but then why not just sterilize the grain in a proper autoclaveable bag?  You can certainly take grain and transfer it into a bag like you want to do just be careful because the outside of the bag wont be sterile.  When they make ziplocks and such they are sterile on the inside outta the factory.  So you won't need to worry about the inside.  Its just contams falling off the top.  If youre in front of a hood then it wont be a problem.  If youre in a GB then it may make a difference.

when I said improved I meant im-proved.  Like winging it, not improving what we already do. Sorry


and to answer the first question, you need gas exchange, not air exchange for colonization.  With that said, use a filter patch bag and sterilize you grain in it.

your environment is not that clean.. trust me...

bags work great, but you need to PC the seed IN THE BAG.. there are bags for this sold by supporters of this site. mycofactory is where i get mine...

there are also teks for that.. anyone got one off the top of their head?

So I colonized 4 jars of GT about 2 weeks ago. I had 2 of them with just the hole in the micro pore that the syringe went through and 2 I took the tape off of the holes. The 2 without the tapes on the holes are way ahead, one of them is over 50% ahead of the others. Could be genetics but only the two I allowed maximum air exchange took the lead. Doing a similar test with EQ's but I would say yes.
