thats not the best way. It may work, but the prob. is that when you shake it, the grain can touch the tyvek, which is bad. I make for nail size holes in my lid, and tape the tyvek on top. RTV'ing it on top is popular

How are you planning inject your spores/lc? If you injct through tyvek it will leave a hole big enough for contams to get in. But if you do you can cover it somehow with micropore tape.

I use the band to hold the tyvek on and when I innoc, I simply quickly lift the tyvek and go in the hole, while using a still air box. works fine.

I'm not using lc or spores. I'll be doing an agar to grain transfer. The grain can't touch the tvek? Because the only way the jars fit in my PC are sideways... what is RTVing? I thought the more GE the better, 4 nail holes is enough?

Thats fine. You probably have a flowhood or glovebox then

Glovebox for now. Once I get into a house i'm going to try and build a flowhood, or just buy one.

yes, the grain touching the tyvek is bad. If there is just a drop of water on the grain and it gets the tyvek wet, with nutrient-enriched water, bacteria can germinate on the outside of the tyvek and grow right through it. 4 tiny nail holes is plenty. RTV is high-temp. silicone caulking you make automotive gaskets out of, it is usually orange. check out the whatman lid tek

With a single layer of Tyvek, you can easily have too much. A large G/E hole can allow the jars to dry out.

So should I double it up since the holes are so big?

@Randy,
I did what you did for my initial batch, and guess I got lucky but I definitely saw the flaw in having the Tyvek layer underneath the metal lid. I use just polyfil now, but if I were to do the Tyvek layer, I would do Jar, Lid, then Tyvek.
If you do Jar, Tyvek then Lid, moisture gets trapped between the top of the tyvek and the lid, which is bad. Also because the entire tyvek layer comes into contact with the bottom of the tyvek when you shake it is another vector for contam.
Also I have 4 nail holes on the sides and one center hole for GE. The 4 small holes are covered with masking tape during the PC. I inserted the needle past the tape and tyvek and then retaped it when I was done.

It really depends on the RH of your surrounding environment and how wet the grain will be. You might try it on a jar and see how it turns out.
