How often do you change your Agar Recipe?
    #14311954 - 04/18/11 01:13 PM (12 years, 10 months ago)

I know its beneficial to change up an agar recipe so that the mycelium gets a change of pace and doesnt get too acclimated to one food source, but how often should the recipe be changed? Should it be everytime that a transfer is made it is onto a new agar recipe?

One other question is what are some easy recipes that you like to use? I have only tried malt extract yeast agar (MEYA). I have some dextrose that i got from a brewers supply, can i substitute the yeast with dextrose as a different food source?

what you're talking about is probably called culture senescence and I think I've read it varies from specie to specie but is a number around 120 transfers.

so your agar recipe is safe for one hundred transfers at least I'd say, which is more than enough for most of us

I change mine every transfer.
Not because its the best.
But because it is easy for me.

I have a bottle that holds enough agar
for 20 plates. and each sleeve has 20 plates
so I simple mix up a different recipe each
time i pour some plates.

I only use 2 recipes

PD(Y)A  &    ME(Y)A

some times i do not use the yeast
It depends if I have any on hand or not.
And I have had good success doing this.

Some people don't change there recipes
and some people do. you will hear different
experiences on what to do and what not to.

I find it beneficial to change the recipe
as to how often to change it... I don't know

it would be save for a thousand of transfers.

if you were a hungry mycelium, would you refuse food one gives to you, if nothing else is available?

the mycelium wont get sick of eating the same over and over again.

cubensis mycelium is not a gourmet; it will eat nearly everything you serve.

That's only a theory and not one I subscribe to.

I've had all of my cultures on MEA for well over a decade, and many of those were more than ten years old when I got them.  I've not noticed one whit of deterioration in performance.

MEA is the easiest agar recipe available, IMHO and is what I use.  Sometimes with the addition of mycological peptone or brewers yeast at .5 of a gram every 500ml of water.  MEA is just 10 grams of LME and 9 grams of powdered agar agar per every 500ml of water.

I wouldn't substitute brewers yeast with dextrose.  Dextrose is a pure form of sugar and brewers yeast is yeast.  I'v also never read of an agar recipe calling for dextrose.  I'd just keep the brewers yeast for use with agar and the dextrose for use with LC's.
