i never do grain masters. do some search but are too many results and is too much confusing

I did plenty of G2G in a glove box before building my hood. A flow hood just makes the entire process easier.

I prefer taking a syringe and shooting 1 cc into a jar..then split that jar into 10, then each of those jars into 10..then you have 100 jars from 1 cc, and all the genetics are about the same.
You can put 1 cc of spores into a LC then use that to make 100 or 1000 jars the same, but it doesnt seem to work out as well as G2G transfers.

I like to go spores to agar, to grains, then expand the culture on grains from there with grain to grain transfers. I think LC's can add unnecessary vectors for contamination, require extra bells, whistles, and maintenance (syringes, stirbars/plates, jars, whatman filters, extra jars, lid construction, and syringe maintenance, ect..).
LC's are an all or nothing gamble and are best assumed contaminated until test substrates or mediums have been inoculated and have had 5 days to come back contam free. Gambling can be fun but in all the time it takes for LC's to colonize their medium and be tested for cleanliness, you can have several jars that you know are colonized without contamination ready to be expanded into what ever (bulk substrate or sterilized grain or substrate mediums).
