If you were to run an experiment to compare two bulk substrates how would you choose to do it?
Compare biological efficiency by measuring the dry masses of the sub ingredients versus dry yield? Over how many flushes tho, 2 maybe?
What spawn ratio would you test first?
How many runs would u consider significant?

Substrates are usually sold by the square foot or inches. You would have to take into consideration the cost of the substrate vs the production ability to see which is the most efficient. I would do 2-5 tubs of one thing and the same amount of another. Measure out each tub dose by volumetric proportions and basing it's bio/cost efficiency on yield. You would have to do multiple tubs to limit any real variables. Sounds like an interesting experiment though.
EDIT: I'm sure you can come up with a mathematical equation to assist in the judgment. SxC/W Substrate Volume X Cost / Dry Weight in Grams or something to the effect.

You'd also have to isolate on agar and use the same isolate for each tub, otherwise differences could be due to genetics instead of the substrate.

Oh yeah, I forgot about this too... Say one bag of substrate is 6sqft and cost you 34.27 and the other bag was 4sqft at 19.33. You'd have to figure out the cost of each ft to judge the price.
3427/6=571 (5.71/ft) 1933/4=483 (4.83/ft)
Obviously bag 2 being more cost efficient. Lots of variables need covering.

I wasn't going to get into the cost comparisons yet, but that certainly is an element of it.
Manure is free, coir is not, but manure has more labor involved. I get wbs for .20 cents a pound. This might imply using a higher spawn ratio would be beneficial (since it's cheap), but the more spawn I use, the longer it's gonna keep putting out flushes. Considering the value of fruiting space I'd ideally want to dispose of the sub after the 2nd flush, maybe 3rd?
So yeah, figuring cost efficiency is exceedingly complicated. I'll just start with looking at actual performance without reference to cost.
