Analysis of colonization problem
    #1424736 - 04/02/03 10:41 PM (21 years, 6 hours ago)

That damn amphibian just won't leave me alone. This time, he asked me for advice on why his jars had neither colonized nor become visibly contaminated after 14+ days. Since I don't know the first thing about mycology, I thought I might refer the problem to you and pass the info on to him (lousy amphibians, mooching the internet and refusing to drop any dough). Could his jars not have had enough water? Or did he kill the spores by flame sterilizing the needle? oR any other suggestions so he wouldn't repeat the mistake?

Substrate was most likely too hot during innculation, causing death

We don't have enough information to validate this.

What type of recipe did you use? What type of sterilization method?

Standard MMGG substrate, pint jars, he let the needle cool for about 10-15 seconds after sterilization, and no alcohol was used to help sterilize.

your needle may still have been to hot so the spores died when going through the needle. Thats what i think. how about everyone else.

I've innoculated jars with a hot ass needle with no problem..the first drops of water cool down the needle. So I doubt that was the problem. I'm not sure that too hot jars was the problem but that's my best guess based on the information given.

Ya..either bunk syringe.. jars were too hot.. or something else I can't think of.. how long did you let the jars cool after you cooked them?
