To poke or not to poke

I'm still a newbie to all this.. I'm on my second growing batch and my first cakes went great. When I was preparing the substrate for my first batch, I made the mistake of putting them in the pressure cooker without any holes poked in the lid (I'm using canning lids, not foil) I went ahead and hammered 4 holes in each lid, after wiping the top off with alcohol and doing the same to the nail. You could hear the air being sucked in. All 5 of my first jars inoculated fine with no contamination and my second batch is halfway colonized and looks great as well.

My question is- Does it really make any difference if the holes are poked before or after the sterilization process?

it's better if you poke the holes to prevent the lost of moisture...

Of course, it only matters the first time for each jar ;-)
But before is generally better, as you can take them off the jar, poke the holes, then put it back on, then cover them with tape.
Glad you were lucky and didn't get contams - since as you noted, it sucked a bunch of air into them when you did that. That's the big reason you don't want to do it afterwards.
