any flaw in basic spore/syringe

Hi I was just wondering whether anyone could give me any advise or any mistakes I should avoid, TIPS. I am going about making a spore print and in turn a syringe.
could I place a cap right into a alcohol wiped little tiny rubber maid container or even a small zip-lock bag. Using no foil, just let the cap sit right on the zip-lock bag and or tiny rubber maid container? the placing of the cap into the Ziploc or rubber maid would be done in a glove box.
Once the spores had dropped could I just open the container or zip lock bag, IN THE GLOVE BOX, take out the cap and with clean syringe shoot the water in the container or zip lock bag and mix and suck up the water. I
Or Would it be a better idea to have a cap sit on a sterile foil in a sterile zip-lock bag,in a a glove box.I would then take out the cap from zip lock bag, leave in the foil with the print and just shoot in sterile water from a few syringes right into the bag with the foil still in the zip lock bag or conatiner. Mix it around with my clean gloved hands in the glove box. Should I then poke a hole using syringe into zip lock bag to suck up the water and spores or just slightly open the zip lock bag and stick in the syringe to pull out the spores and water? same goes could I do this with a rubber maid container? Thank you so much this will help me avoid making a big mistake with innoculating like 14 jars using one cap

I like that you actually care to think these things through but, honestly, you may be overthinking things a tad. On foil in a glovebox/mason jar with lid/upside down container OR in a plastic bag with no foil will suffice.
With the foil method its as easy as (inside a glovebox) scraping the spores into a jar/glass of some sort, injecting sterile water into said jar/glass, stirring, aspirating spore laden water and voila. Some like to shoot the solution back into the glass a second time before aspirating a final time but I've never found this step necessary.
With the bag you simply place cap in bag, let it do it's thing, remove cap (using the same sterility you would to remove it from the foil), seal bag up, shoot water into bag, mix, aspirate and donezo.
Gloveboxes/flowhoods are recommended all steps in the process of making a spore syringe.
Oh and you'll get waaaaaaaaaay more than 14jars from 1 cap dude. 1 print goes a long way.

stick a tack or needle etc in the cap,, stick it in a clean 1/2 pint jar on the bottom put lid back on and let spores drop for 18 hours,, remove cap add sterile water the suck it up with a sterile needle,,, all in a glovebox or flow hood for best results!

awesome thanks alot guys. I think I will do just that. I will take a sterile needle to cap, chop off cap with sterile scissors. place cap in a small sterile mason jar, let cap sit right directly on bottom of the jar (no foil). I will have made a small hole in my mason jar lid that will be covered with tape while spores are falling in the jar. The jar will be in a glove box the whole time. When its time I will take out the cap. cover jar back with lid that has a hole, take off tape from the hole, and mix my sterile water and suck up using the hole in the lid. i think the hole is just another safe guard instead of exposing the whole jar ( no lid) while sucking up spore water. Think this about covers it. Thanks alot

Remember though, the cap itself will not be stirle, but you still need to take the necessary steps to keep from getting any extra contams.

if I place cap directly in a sterile zip lock bag after cutting. bring it into glove box, and take cap out and place in jar, the cap will not be sterile? it can contam the jar ?? any precautions I can use from cutting, transferring the cap into my glove box? Thanks so much for your help makes things easier, i am always full of questions, I have been searching the forums for hours this past month but sometimes its hard to find specific answers

RR recomends iodine but I don't know the specifics as I am a noob also. I do know that he does NOT recomend peroxide. Try searching "making a print" P.S. Anything RR or Roadkill says is word, they have been at it for a long time.
