is this an isolate?

Howdy folks. I guess I have a few questions on these plates. Let me begin by giving some background. I have recently been trying to find a viable way for long term storage of genetics. I figured what better way to do this, than to first come up with an isolate, with alot of rhizo growth. So I first took some PE spores, and inoculated grain. I grew that grain out, and spawned it to horse manure. Then I fruited out the sub, and took a rather large fruit, in a cluster,and cloned it to agar. So at this point, I have done 2 transfers on agar.My understanding of how to determine, if I have achieved an isolate or not, would depend solely on the growth not sectoring. I have made 9 dishes up, from one master plate. I tried to pick the sector, from the master plate,with the most rhizo growth.Also these plates are 9 days old. here are my questions.

1) If all 9 dishes are from the same sector, why dont they grow the same? why am I seeing sectors on some dishes, but none on others?

2)why am I not getting alot of rhizo growth, If the sector I took from, was very rhizo? Could it be that they are still a little too young?

3) do any of these dishes appear to be an isolate?

4) dose my strategy for attaining an isolate, seem correct?

Ya I know, they are a little rough, but its the humidity on the lid of the dishes.I wish they wouldnt do that,but I can see a few of them pretty good. see dish 4 and 8.
