What do you think? Do you see sectors, or is it an isolate?

I don't see any, but take pictures of the other side too to make sure. RR

Pics from the other side didn't come out well, but to the eye it looks about the same.
Thanks for the input!
Edit- oh, I just noticed a contam spot! It was a sloppy pour, I'm lucky to have got what I did. Transfers have been made.

how many transferes was that?
i really wanna invest in some agar. ive tried doing the same concept on a 1 inch layer of brf in a jar. i was able to get growth, but they got really fluffy instead of ryzo. sorta like the fuzziness on the stems of mushrooms lacking either humidity or fae (im not sure which)

Five or six transfers away from a tissue clone. Probably about two dozen plates total to get that. Buy agar locally, and you can use PF jars as dishes. Short term and small scale, anyway.

when making the transfers, should i construct a glove box. or can i do this in open air if i clean a small room and bleach the hell out of it. Asian groceries usually have this stuff right. is it labled as agar or does it have another name.

With a surgical facemask and gloves, you could probably get away with open air inoculation. But, make and use a glovebox for better results.
Agar flakes, agar powder, agar agar. Available from asian markets and health food stores. Pronounced "auger-auger".

Shoulda had 'bacteria' as an additional poll option
I don't really see any sectors there, but neither do they look as uniform as I'd like them to be before slanting. I reckon I'd do one more transfer into 3 individual dishes and let them grow out a bit to be more confident.

Doc T I don't have a clue but I'm wondering about the piece above your thumb at say 7 oclock. Why is that hairly while the other areas are coarse?

It's condensation and/or the plastic bag making it look fuzzy.

I don't see any sectors, but they sure look rhizomorphic.
Did you just do that last transfer because of the contam, or for reassurance on whether it is an isolate or not?

You're closer than you think. Go get some agar and get ready.
Final transfer to confirm no more sectoring and then those will be my working jars. This one is in the fridge right now, I'll get it out tomorrow and remove the whole slice where the bad spot is.

As this looks like it is not partitioned, what will happen when all 3 mingle with each other??

Doc_T, how often do you transfer?

If it's an isolate it won't matter. And it'll be in the fridge anyway, not growing.

Once a week or so? As needed, I guess. Whenever the myc gets about an inch across or when I see a sector I like.
I incubate agar dishes at 78-80F btw. And when I do agar->grain transfers, those jars go in the incubator at least until the first shake.
