Been cautious man, I though I would start with a smallest, smallest quantity. Namely I started with 0.25mg.....and my intention was to increase dosage to see if something will happened.
That very small dosage, the first dose of my experiments i planed, was very very strong.

i cant tell ya from experience, but they appear to look like they're 'browning' like right before you get ready to fruit blocks and such...idk what healthy shiitake mycelium looks like other then on agar, and in LC

Looks like you should have spawned those a while ago.

Also I would suggest removing the tinfoil off your jars after PCing
to provide better GE.

And what about contamination rate? Isn't better with foil?

I've been growing shiitake in jars and bags for almost 2 years now. I live in south africa and its almost impossible to get the micron air filters down here so Ive resorted to other means of preventing the spread of contamination. Foil is really the only option if you don't have the filters so keep on using it if you can't find any. Cable ties are also great to use around the foil to ensure that no ants or bugs decide to march under the foil and contaminate your agar or substrate

The browning is caused by oxidation of the surface of the mycelium. Its a natural evolution of the fungus as it naturally grows on logs and spots which are in direct contact with air often turn brown. If you dont know, its a natural defense mechanism of the shiitake mycelium and any contamination or mould which occurs after browning will do very little to the yield of shiitakes that you will get from the block - other than the nutrients which has been consumed by the foreign contaminant, although the shiitake often just consumes the contaminant as well. Shiitake is the "old veteran" when it comes to cultivation as it will eventually kill almost all contamination that appears if it is well enough established.

Oxidized mycelium cannot be used to inoculate another jar successfully. The yields fail and the rate at which it colonizes the new substrate are really really slow by comparison. If possible I suggest bringing these to fruit and then take some fruiting body tissue and put it on agar to restart the growth of your mycelium. This revitalizes the mycelium and causes it to grow faster and produce higher yields. Its suggested that this be done at least once a month to ensure the growth speed of the mycelium.

What I suggest is that you mix a solution of water and hydrogen peroxide, 3% concentration of 50% strength and then let your jars soak in that overnight in the fridge to cold shock the mycelium. The hydrogen peroxide causes bubbles to form on the outside of the mycelium against the glass and this often is enough to cause it to come loose from the jar. After it comes loose let it drain and take the grain out inside your laminar hood or glove box and move them to the fruiting environment.

If they do not fruit, cold shock them again in distilled water or hydrogen peroxide solution and try to fruit them again. If the block still refuses to make a fruiting body your best bet will be to break open the block inside your glove box and use one or 2 of the grain kernels to inoculate another jar of substrate which you can then bring through to fruiting. TAKE NOTE THAT THE KERNELS YOU USE MUST BE COVERED WITH WHITE MYCELIUM OR YOU WILL BE BLEAK AT THE RESULT AND THE TIME IT TAKES TO GET THE SHITTY RESULT!
