Ive an Autoclave similar to a 41.5 quart all american pressure cooker and i use it to sterilize my saw dust. How long normally must one cook at 15 psi to sterilze the substrates?

For supplemented sawdust I usually do 2 hours at a min.

I never do longer than an hour, grains or saw, supplemented or not.  Rarely have problems.  I don't start the timer until it taps and I don't take the lid off until its cool to the touch.

The volume of the containers you put in the PC will dictate the cook time really. Large gusseted bags will need more time to heat and sterilise through than jars.

well, mine is not supplemented just saw dust and calcium carbonate and calcium sulphate with water. I intend to use grain spawn once it cools.
My mini-trial of just 40mins after 15psi proved successful. Any others have the same result?

And i put my substrate in the drum and pour in pail, as where im from, they do not have autoclavable bags.

Also, i intend to do a trail with powdered supplement to check the differences between grain and powder, Anyone else does so? And cooks for how long?

I believe 40 mins would be successful.  Just likely not to be as successful over the long haul as 60 minutes.  People tell me all the time that 90 is significantly better than 60.  If I ever experience that in my env, I'll switch.  Really, your environment has a tremendous lot to do with these factors.

Depends on the size of the blocks you're using and how packed the cooker is.

Those are very good ans. In short, one has to try on his/her own through trial to establish what works and what does not.

Also, can one load a dry substrate and then pressure cook as water is already added in the pressure cooker. Once cooked, just release the water outlet and not the drying outlet. For i believe, as water boils and steam is circultaed all around, the sawdust is already wet.

What about the moisture content? HAs anyone tried this before i do?

Adding dry substrate won't work, the stuff won't hydrate much, when steam sterilized.
