inoculating agar with syringe...

Just wonderiing if I would Just place a few drops of spore water in the center of the agar or would I do a streaking pattern like with an innoculating loop... or doesnt it matter??

try dropping a drop or 2 in several spots...at least 3....generaly agar holds enough water to germinate the spores but if a syringe is your only source...well do what ya gotta....

multiple inoc points help it to colonize faster and more completely....plus you have a better chance of a clean culture

Well the only reason I will be using a syringe is because they are old spores and I figure that the spores will need to be rehydrated before I inoculate the agar...

What If I just squirt a big zigzag of sporewater in the dish.. hehe

But you say the agar holds enough water??.... hmmm

personally, I'd do it in the centre, easier to catch rhizomorphic sectors. Depends how dilute your syringe is I suppose. If it's good and thick, I'd inoculate in the middle, then pick out the sector that reaches the edge of the petri first, then do a couple of transfers to make sure....

Also this is still slightly on topic..

If I were to have a definitely unsterile spore print and I wanted to isolate a pure culture would I :

inoculate as per usual...
Cut out any decent mycelial growth
transfer to peroxidated agar
continue transfers until a pure strain remains..

Has anybody got a good online link for proper streaking techniques to help isolate contams from the inoculant??

You could use peroxidated agar from the get-go and just drop the spores on a cardboard disc in the center of the plate. This will let the spores germinate before reaching the peroxide. Cardboard is pretty selective, so nothing but mycelium will grow on it.
