Hello all. I just got a few spore prints in the mail for Z strain and 2 other strains from one of the coolest people I have met on this site. I told him that since he sent me these prints I would keep a grow thread with my results... Since I am a man of my word, here we go.
I have never done Z strain before, and since I hear it is the most aggressive colonizer out there, I decided to inoculate 4 different types of jars just to see what it really is capable of -
5 BRF PF cakes 3 quart jars of soaked and simmered rye berries. 3 1/4 pint jars of honey based LC. (and when i get the rest of the supplies) 3 agar plates.
- Last night i made 1 spore syringe and did the 5 BRF cakes and 3 1/4 pint LC jars and they are all now sitting in my incubator at 76-78 degrees Fahrenheit. - Tonight or Tomorrow I will be making another syringe for the rye jars and hopefully in the next few days I will start the agar plates. - I will trying to be keeping this thread regularly updated and if anything cool starts happening (or when all my jars are colonized) I will start adding pics... Wish me luck .

I would start with the LC and the agar.

dont incubate your jars.. unless you are freezing to DEATH in your house, you dont need to incubate them.. my jars went from start to finish in 14 days from spore, in a VERY cold house.. no incubation needed whatsoever..

ya i know, people keep telling me this... but with even only a 6-8 degree F difference i notice a HUGE time difference between the ones i put in my incubator and the ones i leave say in my cupboard. i did a rye comparison with treasure coast spores and put 3 in cupboard and 3 in the incubator and the incubated jars showed anywhere from 1.5-2x the colonization rates of those in the cupboard... i know that the increased heat can become a contam risk, but i dont think its a big deal if i keep it below 80.. right?

if you are inoculating all of those jars from multispore and not an isolate, then it wont be a really valid experiment, as every squirt from the syringe could have different results..
incubating also causes sweating of the substrate, which can lead to pooling water in the jars.. plus, an incubator takes up space, is an extra step, etc etc..
feel free to use it.. its just not really needed or recommended.. unless your pee is freezing before it hits the toilet.. haha
