Agar contamination timeline

So i got some culture tubes, that come sterile, but dont have autoclavable caps. So instead of scrapping them, i made a jar of agar with an injection port and sucked the agar out of it with a sterile syringe and filled the tubes in front of my flow hood. I just feel like this adds too many contamination steps in the whole process....
If they are going to contaminate, how long would it take to show on average? 4-6 days?

i've had trich show itself in as quick as 1.5 days, but normally 3-4 days if trich is there its going to show itself.
what I would of done would be, mix the agar, in a pot ont he stove, then pour into each container, then put foil over for a lid...sterilize, and then when you take it out slant it how you want to cool, and put a rubberband over the foil to keep it secure

Here are the tubes i used.... Didnt want to use foil, and i want an airtight seal...

sorry when i read about the caps i thought you said they didn't have caps, not that they weren't autoclaveable, my fault...thats the only reason i suggested foil!

You could always put the caps in front of the flowhood and pc the tubes with foil, then open your pc in front of the hood and put the caps on...possibly? Even if the pc is huge, you can let them cool in it and put it on a chair that's just lower than the surface the filter is on.

Ill let you know how it works out... Ive been having alot of problems with my agar recipes anyways...so who knows if anything will grow... Only thing iv got growing on agar lately is polypores.... Maybe PH?

What recipe are you using? I found (by eyeballing) that slightly less nutrient agar seems to colonize faster, but that could also be the strain. If it contaminates youll know within a couple days. Best of luck on it!!

10g malt 9g agar 500ml water
I think the past i wasnt using distilled water, so there might have been some bad ph... I think(hope) these work out better

I use less malt than that and it colonized pretty quick, but it could just be my curse, and I've always used tap water. Shouldn't be anything wrong with the ph I wouldn't think unless your water is high. I'm no expert and don't know what exactly it could be, but from my experience a little less malt would be my suggestion.
