LC is slowly growing from tissue sample... I hope!

I have lurked for a very long time and gotten so much wonderful information from this forum! Many thanks to all. However, I have a question I haven't been able to find an answer for, and I'd like to participate regardless. So here goes:

Loosely following creamcorn's clone LC tek, I cut a tissue sample from inside the base of a mature mushroom and placed it into a 4% Karo LC. Nearly two weeks later, the solution is still completely clear (awesome), and there is a small white puff around the sample. I've included a picture, but my iPhone is not great at macro photography. This looks promising, but according to creamcorn's tek, I should be fully colonized by now, whereas this hasn't moved much at all in the last few days. Room temperature is in the mid 60s. Some say to incubate, but I recall RR saying that it just encourages contaminants and doesn't help mycelium, so I haven't thus far.

Should I just wait it out, or should I change something?

have u been shaking it? if not now that it is showing growth i would shake it as often as posable.

and its almost impossable to judge how long it will take for lc to collinize expecualy from a tissue sample. ive had some lc's colinize in a week some took a month.

Responses so far are very encouraging! I swirl it every couple days, but the puff cloud stays with the tissue as one unit. In case the pic isn't giving a clear picture, the cloud is a little less than an inch in diameter, and there's nothing else in the surrounding solution.

yeah you should have something in there to break the mycelium up when you swirl it..its going to be hard to suck it up if ya cant get it broken up.

Hmm, that doesn't sound good. I've read that sucking up mycelium with a syringe and forcefully expelling it can break it up, but I'm worried about introducing contams while most of the jar is uncolonized. Is this something I would need to do soon, or can I wait until the mycelium cloud has grown significantly?

using a syringe like you said should work, but
keep in mind that the myc is very thick and it
easily clogs an 18 guage needle.
you could make a second lc WITH a marble or
something and shoot whatever myc you
CAN suck up into that.

Ahh, good call. Ok, I think what I'll do is let this one keep growing all it wants, knowing that it may become unusable because of being too thick. Then at some point I'll suck up as much as I can and use it to make a new LC.

at least once/day give it a good swirl. getting some air moving through the liquid promotes growth as does spreading out the mycelium so that more points are exposed to the nutrients in your LC.
