Growing from very light spore print ...
    #14133697 - 03/16/11 09:10 PM (12 years, 10 months ago)

Hi, I'm looking for the best way to get some spores onto agar from a very light spore print.

From my last Oyster grow I made a couple of spore prints. For some crazy reason I picked a couple of very small caps (~1in) to put on foil. They may have been too dry or I may have missed the drop but they turned out very light - barely discernible (~1/4in spot).

Now, just for shits-n-giggles, I would like to try to see what would grow from these spores. I need to get them on agar and have a few ideas and would like some feedback.

Should I:

A: Put a drop of sterile water on the foil with the spores with a dropper, mix it around with the dropper, then gather what I can with the dropper and put it on a plate(s).

B: Put a drop of sterile water on the foil with the spores with a loop, mix it around with the loop, then gather what I can with the loop and put it on a plate(s).

C: Cool a loop on agar, smear the tiny amount of agar onto the spores, then gather what I can with the loop and put it on a plate(s).

Please offer any other options.

I've even considered after having using method C, placing the foil - with the agar and spore remnants, into a empty, sterile petri dish to see if anything will grow, then transfer that to a plate.

Again, I have viable cultures, this is just to see Nature's determination. I want to see if I can get a very small amount of spores to fruit.

I would go with C.  You don't need a large number of spores to get a culture to germinate.  And the fewer tools and stuff involved the less places to get contaminates.
