I'll probably sterilize my enoc loop and use that to carfuly scrap some mycelium into the tub of water.

I take my scalpel, scrape some mycelium off the plate, JUST MYCELIUM, and put in the vial then shake it hard and store with parafilm around the cap.

when you scrap it, do you hold the plate over the vial or grab up as much mycelium as possible and put it into the vial? If you do number to I assume you don't have any part of the scalpel touch the interior? Do you sterilize your scalpel along with everything else and then sanitize with alcohol and flame?

I am sorry to have been confusing. What I ment to say is that I soak the tooth picks in a malt extract solution in a small beaker and then pc on the same run, with a batch of agar. I would not run the pc just for toothpicks, I try to make my runs count as every one cost in overhead. The agar and the toothpicks are a separate media.
I usually start the week with a "tissue day" extra clean lab, pour agar, make plates, make slants, sometimes g1, then the rest of the week is cooking wood for inoculation. I like to start my week with some agar work just so I easily keep up on making cultures, and it is sometimes a half day, but usually I do agar then go cook wood for the first batch of the week.

so do you add the tooth picks that have been soaking and pc'd in the ME solution to the agar later? I'm just confused with the tooth picks being separate from the agar.

tooth pics alone, in a small beaker submerged in 10% ME. covered in tinfoil. Drain water on flow bench, use tweezers to transfer into inoculated slant.
The agar has nothing to do with the procedure. I was just commenting that the toothpicks Can share an autoclave session with things like agar, or lc media etc.

ya I understood that you where saying that it could share a PC session, but wasn't really understanding the rest. So you transfer these to a tube of myceliated agar and they grow on the tooth picks and then you can use them to enoculate am I correct? I also heard of a method involving paper cat litter pellets for storage?

exactly , I inoculate my tube and place the tooth pic with one end touching the wedge and the other just in tweezers reach of just inside the neck. The agar "adheres" the toothpick in place. They are kept horizontal to grow a little, and once your myc grabs the toothpick and the agar base, they go into the cold.

your hood is bigger than mine, I lay the bag horizontal and open the top resting against the sealer. I inoculate fold the bag over and seal. I have done hundreds of bags and had very little contaminant

thanks... got everything ready to do my slants today, now I just got to get these damn tubs to fruit... been 3 days and no knotts yet. the rhizo's are completely vertical and so thick I could cut them? Hope I get some mushies soon, i'm getting anxious. want to eat some

I figured since i just did some, and its relevant, ill post mine in this thread! These are not the first i've done... Did some in glass vacutainers , but i dont consider them, as they have never been used past a trial...
Pictured below are the slants...Sorry for the horrible quality, my macro mode sucks...
   
Some information on the tubes... I received some cheap pre-sterilized PP tubes and caps from cynmar. They were advertised as all PP, but turns out the cap wasnt and melts during autoclaving...  Anyways, to make use of a product other wise laying around in my lab, i sterilized some agar and filled them infront of the hood with a sterilized syringe... Cooled them at a slant and then inoculated them with a few iso's around the house.... Wasnt hoping for great results, but got some pretty decent ones...

my pinning is strategy is OH I left these in the bag too long. I have a clean room full of bags. I rack them like wine. I put them up and then forget about them, and then put them out or apply them when and where they will do the most good.
I maily produce spawn for other people. Most of my cultivation efforts are along the same lines. Make a bunch of spawn, put out a patch somewhere and forget about them until they show back up.
