subtrate formulation and isolation

is it better to keep your agar substrate the same to keep the mycelium running durring isolation and once completed then switch substrate types to develope other digestive enzymes?

I read a post where RR claimed he has never changed from using MEA for his cultures.

I've just not found evidence of the 'digestive enzyme' training phenomena.  Honestly, if I had I'd have switched agar formulas around or switched away from rye berries, but it just hasn't been a problem, at least with the species I grow.

thanks RR.. I have noticed that certain of my strains perform better on MEA while others better on PDYA.
I bought some lab grade crap from a local supply store and it contains the following per liter,

15g agar
20g malt
20g dextros
6g pepton

way too much, noticed that it made the mycelium very lazy and seems to promote mutation factors..
I just made a batch of sweet potato agar and am hoping that the complex carbs will promote some good growth.

it does depend on the species as well, I have not been able to keep an Agaricus spp on PDA for more than 4 transfers and it starts to show sectors and lose vigour.

But oysters do not seem to be bothered.

ya oyster seems to like the PDA better but then the my cubies seem to like the MEA slightly better as long as the sugar content is lower.
Thats why I'm hoping that the cultures will responde well to the sweet potato agar that I made 2 days ago.

ok so two things I've noted. 1) so far the sweet potato agar is already showing signs of thick rhizomorphic activity.. the mycelium is gearing to explode.
2) what I've noticed with nutritionaly dense mediums growth is very slow and seems to promote mutation, or more so accentuate the different strains on the plate without allowing the mycelium to grow over a large area of the plate in a short time which would in theory keep the cell divisions down to a minimum.
So basicaly say my plate grew less than the size of a us quarter and showed def sign of separation, i would be able to pick and isolate my mycelium without it ever expanding significantly.. seems that alot of time when agar isn't as dense the mycelium tends to outrun the plate befor you are able to pick up on it's true nature, because its so franticaly searching for nutrients.. def something worth looking into, but anyway as of right now the sweet potato agar is rocking and showing alot of promise.

Yea, that's some healthy looking myc. Might be fun to try that with a few other types of roots/tubers... taro, yucca are a couple I can think of atm. Would be a fun little project
