time for some fun!

So here's what I have:

4- Quart Jars WBS
4- Quart Jars Rye
12- 1/2 Pint Jars PF tek
4- 1 Pint jars with Karo/Water LC
and 10- (empty..like, no agar) petri dishes

all waiting from some MS syringes to get here in the mail. which should be in the next week or so!

Seeing as how this is the last effort I make in my mushie adventures (moving cross country in 5 weeks...) in awhile, I want to try several different things at once and see what happens!

I'm thinking MS inoculation on 2 of the Rye and 2 of the WBS jars. MS 'noc on one of the LC jars. Mix up some Agar and get at least 4 or 5 of my petri's rocking. Take Agar cuts and 'noc up one WBS and Rye jar and the remaining 3 LC's, and once those colonize well enough use the LC's to 'noc up the remainging Rye and WBS jar! and of course the PF tek I'll probably just MS 'noc them all. Unless anyone is interested in how fast an LC would take over one of those things..

wow that gets more confusing every time I read it. Anyways, what do ya'll think? anything else you guys would like to see? Also, I have plastic petri dishes, does anyone know a good tek for setting them up with agar? I'm afraid of their durability when it comes to my pressure cooker..

Are you planning to get from spores to fruits in 5 weeks?

Also are the petri dish's wrapped in a plastic bag or out loose?

well if I could get fruits in 5 weeks or so that'd be NICE. But I'm not naive enough to EXPECT those kinds of results. Whatever I have on hand that is going strong but isn't quite ready to fruit I'm going to give to a friend of mine. :smile:

The petri's came from a lab with agar all wrapped in plastic. I knocked up 4 with some LC jars that I had to see if they were contaminated (yes, they were.. lol). I dropped a few pieces of grain from a monotub I had at the time into a few more (I've never messed with agar, so I just started culturing about anything I had on hand haha) and the rest contaminated by themselves, probably because I didn't keep them in the fridge. oops.

But I have since washed them all out with hot water, antibacterial soap, and then gave them a good rub with some 99% alcohol. I'll re-sterilize again before I decide to introduce any kind of medium to them.

You could maybe get mushrooms using pf tek.

If the petris are plastic, don't get too optimistic about reusing them.

yeah, that's what I figured.. They were only like $15 or so for a pack of ten. At least I learned something.. I think. :wink:

Disposable petri dish' are just that disposable. I have tried many
ways to save petri dish's and nothing has worked thus far. Even cleaning
them, then wiping down with ISO in front of my flowhood and letting
them dry did not work out.

Are you using a glovebox for inoculating your petri dish's, and what
are you wrapping them with after?

For small scale use, PF jars work well for agar dishes.

Thanks for the links EM666! very good info. Kind of makes me want to build a flow hood however I don't plan on being able to use it for awhile once I move :frown:

I do have like, two Ball 1/2 pint jars. they're not wide mouth, but then again I guess it wouldn't REALLY matter for agar. Any idea where to get agar agar? I've gone to Walmart, Asian markets, and even our local grocery store has an ENTIRE ASIAN FOOD ISLE. Like with stuff that has NO ENGLISH ON THE PACKAGING AT ALL. And they don't have agar agar either =(

Asian market will have it, ask them. You say it like "auger-auger".
Local non-chain health food store will have it too, but it will cost more.

Most common seem to be Telephone Brand or Gold Cup. Or Swallow Globe if you want colored agar.
