might be loosing my strains what should I do?
    #14053837 - 03/02/11 10:37 AM (13 years, 3 days ago)

so I have a fuck ton of plates with about 4 different variaties on it. most of them are in there p-2 phase.. I know that  subculturing 2 times in a row on the same medium is a big no no, but i already had PDYA prepared and had to go out of town for the weekend. also the heat was slightly increased because of the really cold weather here. the cultures are now 12 days old and are on the top shelf in my lab. all plates are going from very rhizomorphic to linear on the end..
Should I be worried?
Is this reversable or too late?
did the possible increase in temp cause the mycelium to exert its self and metabolize quicker?
I can't afford test tubs right now or racks... dont want to loose all my stuff though

Don't worry, two plates of the same medium in a row is nothing.

so whats going on? is this reversable?
Also my Red Boy master dish is knotting up, i've already pulled one mushroom from it and cloned to agar and about to pull another one, also the P-1 plates are showing signes of knotting. thats a good thing, showes vigor but it also worrisome due to it wanting to reproduce very fast...

If it is reversible in your case, I don't know. But I have transferred across multiple plates of the same medium and not noted any loss of vigor or yield. Over many transfers it will though (likely), but two is nothing. Unfortunately, it is very hard to find facts about this, and it varies depending on species as well.

If that was true, I'd have lost all my cultures years ago.  I've used MEA exclusively for dozens of years.  I have many cultures over ten years old and they've never seen anything but MEA, ever.

ok so what should I do? is the temp effecting the mycelium? because it was extremely rhizomorphic and is turning linear on the ends...
I even took the fruit from my one plate and cut it in half and watched it grow into some beautiful rhizo's and it's doing the same thing. the plates are about 3/4 covered. the linear growth is about 2mm or so

Mycelium will often change states back and forth at will.  It seems to have a mind of its own.  If your plates are 3/4 grown out, the mycelium should have already been isolated further or transferred to grains.

yes I noticed growth rings and what seems to be resting phases prior to growth, also the Red boy seems to get it most explosive growth around day 10-12. when is a good time to isolate it and also if you keep moving mycelium from plate to plate how do you get a 3/4 growth in order to enoc grain masters?
I will make some pics and post them,my PE is especialy troublesome

They change growth to better use the substrate you're giving them.  Sometimes you'll get rhizo growth that likes the plates very well, othertimes not.

What matters is what do they do when you put them on grain...?
