My first bulk ain't doing so well, but are my master grain jars ready for g2g innoc?
    #14046746 - 03/01/11 02:49 AM (13 years, 4 days ago)

I started on my first bulk process, but I wasn't as clean as I should have been. Almost all of my jars have completely stalled and I think they are contaminated (I use 1 liter WBS jars for spawn).

However, two smaller jars are the only ones that have almost finished colonization. I have a feeling the syringes I used were contaminated from the LC I made. However, it looks like the myc has won the battle in these 2 smaller ones (500 mls).

I am going to completely sterilize my entire place, and attempt to restart my bulk grow from the 2 colonized jars, doing a G2G into about 15 other 1 liter jars. I will toss th WBS from the jars that have stalled. Question is, do you guys recommend this and do you think the jars are ready for G2G yet? I want to toss my LC away as well, and do a fresh clone from when my bulk grows.

Let me preface by saying that I have never worked with WBS as I have a particular preference for rye grain, however, I have a couple of observations and questions that may help you.

First of all, what makes you believe that your other jars are contaminated?  Are there any visual indicators?  Smell?  Sometimes people will mistake the cottony mycelial growth as a contaminant when compared to a section that is being colonized by rhizomorphic threads of mycelia.

Next, what temperature are you incubating your jars?  Proper temperature (between 76-86 degrees Fahrenheit) for colonization is important.  Stalling is a common reaction to improper or fluctuating temperatures.

Do you have any fresh air/gas exchange filtered holes in the lid of the jars that are colonizing?  This is another potential cause of stalling jars.  As mycelia ages and consumes the nutrient base of it's substrate, it will exude metabolites during the process of colonization in the form of a yellowish/brownish liquid (which has also often been mistaken as contamination).

And finally, to answer your actual question, again noting that I have no experience with WBS and therefore cannot be entirely certain as to whether the jars in your pictures are fully colonized (but they do appear very close to me):
Yes, you can begin to transfer to other jars if there are no signs of contamination.  Be careful during this process, a glovebox is nearly indispensable in this situation as your risk of contamination greatly increases whenever you expose your uncolonized substrate to open air.

If you believe that your liquid culture is contaminated, it is useless and you should do as you suggested and wait for fresh tissue to culture.  Fresh mycelia will nearly always perform better in the hands of newer growers.

In the end, my friend, we will all be together again.

Thanks for the advice! I am following all necessary procedures to make it work.

I just need the thumbs up from a member of the community to tell me that the jars shown are ready for a G2G. They appear about 90% colonized.

wait for g2g transer upon 100% not before. If it stalls beforehand you might be dealing with bacteria.
