Last minute double checking & questions...

I got all my gear a couple days ago and I'm ready to inoculate my spawn bags , but thought I'd go over everything once with you guys to make sure there's no screw ups.
Anywho, so once I've sanitized everything I sterilize my syringes until the needle is glowing red, and then place them in alcohol soaked paper towels until I'm ready to use 'em, correct?
The syringes have giant black globs in them. Should I shake them up or... ?
Also once I sterilize the needles, and they're glowing, about how long until they're cool enough to use?
Lastly I'm shooting up 3LB rye bags. I'm planning on using 5cc's per bag (the store recommended 3-5cc's). 1cc per a corner and 1cc directly in the middle. Anyone see a problem with this?
I think that's it. Oh, wait...
Once they're pregnant I'll be putting them on a shelf with ambient light in a room that's 68F. From what I've read the light won't cause them to grow invitro, but I'm just making sure.

Shake well. Flame, expel a drop, shoot. No storing in towels. And really, alcohol soaked towel plus open flame? Sheesh.
I wouldn't use more than one mL of spore solution per bag- that stuff is potent. Drip a few drops in several places, like you were saying, then just let them them germinate and spread. In general, I think it's better to do bags with g2g vs spores.
Save the rest of the spores, go get a pressure cooker, and get started on agar work. It's easier and cheaper than it may seem.

That's not such a good practice. The paper towel soaked with alcohol is damn clean, but not sterile. It's best to simply heat until glowing read, and then cool down the needle with a VERY small amount of the solution squirted into the needle. Water has a VERY, VERY high specific heat capacity, so not much will be needed to cool it off.

It's up to you, man.

Are you planning on making more than one injection? If so, yeah, I'd see a problem; that increases the chances of a contamination during inoculation 4 times over. Just inject once, and roughly aim at the different spots, wait until about 20% colonization, and then massage the bag to distribute the culture.

There have been times where it did seem light caused in vitro pinning. That's way down the road though, and won't happen until long after full colonization, and in that case, may happen without exposure to light.

lol, they wouldn't have been anywhere near each other... I'm not that crazy . But I'll do what you and faceyneck suggested and skip the paper towel altogether.

It doesn't seem hard at all actually, neither does bulk compared to cakes. I've already bought the agar and I'll get my AA pressure cooker next paycheck, but I'll be cloning not isolating.

Nope, I'll stick it in and move her around.
Thanks guys .

Oh, dude, you are gonna be LOVIN' LIFE once you get that PC!
I went with the AA925 instead of the AA921. It was like $30.00 or so more expensive. Those extra 4 quarts make a world of a difference.
