A microtome is a really cool piece of equipment, but not very practical for mycology. They're usually expensive and require a specimen to be either fixed or frozen, which would deform the mushroom tissue.

It's what they use in a pathology lab to prepare histological slides (from biopsies etc), but first the sample has to be fixed and then embedded in wax so that it's hard enough to cut. The other way is frozen sectioning, which is used when results are needed quickly. After freezing the tissue quality isn't nearly as good though and would destroy mushroom tissue.

I'll have to put some histology micrographs in my gallery some time.

Hey, that was supposed to be our secret.  :lol:  Seriously though, thanks for the credit.  I've used both methods but prefer mine for accuracy.

Also, after calibrating my way I merely hold a ruler up to the monitor and measure the spores, doing the math.  I can measure 30 spores in less than 5 minutes.  The standard way takes far longer.



Do you have a link to your TEK where you use a camera and photo editor to measure spores? Thanks Mr. Mushrooms.
