Ive read about taking clones from the stem, near the base of the mushroom. Though also (of course) from the cap. Does anyone have a prefernce especially if they are looking to isolate a rhizomorphic strain?

I had better results when I took clones from the stem (quicker recovery & less contamns), but I'm sure that it doesn't make any difference in forming of rhizomorphes. The stem and cap tissue are genetically identical, at least from what I know.

I'm defintely looking for quick recovery especially since i plan on using peroxidated MEA. Once the mycelium has recovered (as far as breaking down peroxide) will it retain this every transfer i make? or will i expect a "down-time" ofr each occasion

Is it possible for the mycelium never to recover? I tried to clone some eddibles, and the piece I took was pretty small. They never recovered. :smile:

Yes, often it never recovers. Many cells die when you cut the tissue with hot scalpel.
I had the best results when I cloned the in vitro grown mushrooms - I think it was almost 100% success. Wild mushrooms were a bit tougher and almost a complete disaster with the store bought mushrooms wrapped in plastic. I assume they sell very contaminated shrooms, at least in my area.  :grin:

Keep a small dish of H202 in your work area to cool the scalpel after flaming it.
That way you dont kill the peice you are trying to transfer.  :wink:
