question about agar.

ok so i've been playing a bit with agar, normally i PC at 15 PSI for 20-25 mins
but since i'm only doing a small amount, i want to use my smaller PC, which only goes to 10 PSI
how much longer should i PC to make up for 5 less PSI?

i dont think you can "make it up"
its a temperature difference that is used mainly to kill bacteria that can withstand very hot temps.
For all of my agar, I go 45 min at 15psi

Go with 60-90 minutes at 10 psi or just use your regular PC to do a small amount of agar in at the standard-average sterilizing time and temps..

That isn't necessary. I go as low as 15 without contamination. 45 minutes is way too long.
9 grams agar agar 10 grams light malt dextrose 500 ml tap water
Boil for 5minutes, pour into petri's cover with foil .....
PC for 15-20 minutes @ 15 PSI
I never lose plates and have been doing this forever
The flow hood helps too though ....

You can make up for it. RR uses his barrel steriliser at 0.5 psi(I think thats the psi he uses, I know its not over 1 or 2 though) for a ton of bags it just takes 8 hours.
People around he use pc's that only do 10 or 12 psi, they just have to pc longer. I think I have read for every 5psi less you have to use 50% more time.
I still pc at 15 for 45 minutes.

Professor, what type of petri dishes do you use that survive the PC? The nice glass type or some high quality polystyrene?
I am about to get into agar work (today actually) and am using 1/2 pint jars since I am waiting for my petri order to come in. I ordered disposables and plan to pour in the glovebox in the future.

Cool, thanks for the info. I mixed my agar pretty close to your recipe but simmering agar agar strips won't melt them, they have to come to a rolling boil (or at least they did for me).
The "let it cool overnight" in the PC is probably a good idea to prevent condensation. Like a fool I yanked mine out of the PC after 2 hours and I see some moisture inside on the jar walls. With the polyfill and tyvek it might dry a little though.
Looking forward to getting more scientific with the hobby.

In regards to my mixing/pouring/condensation/general agar technique, Here is an idea that i have not yet tried And this thread details the PE isolation i am working on, with technique included.
I am new to agar, so this is based on about 1 month of experience.

Yea, I blend them sometimes, other times I just simmer them for 5-10 minutes while I prep the PC and Jars and they all dissolve.
Sometimes I use the powder. It just depends if I make it to the co-op or the transworld market. They both have different kinds.

I know Stamets says 45 minutes. He also charges 45 bucks for agar that cost 2 bucks up the street from my house.
I'm saying I cook mine 15-20 and don't have any contamination issues. Take it for a grain of salt bro

PP, i was replying to the OP, not you.
Believe me, i think that 39.95 plus shipping from WA is a LOT to pay for a half pound of agar.
I got it because because of its antibiotic qualities. And because i was already ordering a case of petris from FP.
For my first ever go at agar, I wanted to hedge my bets, and guaranty success.
Making your own is the way to go. It's easy.

ok well i went at 10 PSI for 35 minutes, and have spores on a few trays, and some APE from a masture slant i had on a few...so we shall see how they turned out lol
