Sterile water
    #13987934 - 02/19/11 12:02 AM (13 years, 14 days ago)

Well, im suspecting my bags are contaminated for many reasons and one of it is water. Which way is better?

1- autoclave the water at 15 psi for min 20 mins

or

2- pour filtered water on a clean container and boil for a minimum of 5 mins?

Or does anyone has a much better and more efficient method of sterilizng water?

What kind of water are you using? How long are you sterilizing your substrates? Tap water comes pretty sterile. I wouldn't tie up my autoclave sterilizing water just to sterilize it again after it's added to bulk substrate. I have a feeling the contamination is getting in elsewhere i.e. sterile technique or it's in the spawn.

I suspect it's contaminated LC.

Im using normal tap water.

In a autoclave, once it hits 15psi,after that, is 60 mins enough?

Water heats up very quickly and evenly, it transmits heat extraordinarily well.
So if all you are doing is sterilizing water, 15-20 mins at full pressure is fine.

But I don't understand what you are doing with the sterile water?

Enough for what?

You don't sterilize water.  You mix the water and sawdust or whatever substrate, and then sterilize.  Time of sterilization depends on how large the containers are, how dense the substrate is packed, type of supplementation, etc.

...60 minutes is the minimum. I time 90 minutes for grain jars, and 120 minutes for large bulk substrate bags (> 4lbs), 45 minutes for agar.
