Finally got around to reading all this, nice writups JF. I have a few questions reguarding equipment and use. Iv never used a culture slant before so my first project, to store the isolates I plan on making, will be using yours. Probably a noob question but why layer? Once you layer and cover with tape how do you introduce your culture or was that the agar broth? I followed the link to those microcentrifuge tubes, are these the right ones; the main reason I ask is that the volume is listed as 1.5ml, while in your instructions the first step is to fill the tubes with 1.5cc which would fill that much up. When injecting your culture into these slants do you store one isolated culture on several of these tubes so that you have a lot of backups in case one fails or you want to be able to withdraw several tubes worth(or do you even widraw)? How long do these last. Sorry if its a lot of questions, im too confused today

i layer so i dont cut into the agar, i just suck the stuff up instead the little vials i had were 3cc i think

Oh, so the top layer stays liquid such that you can aspirate, which one contains the genetics? Ill have to look around for some of those vials.

add the agar broth after the agar layer has some mycelium on it or just use an lc to inocculate the agar broth layer
