How do you guys quickly go from isolate to PF cakes for testing?

I've read it said by a few people that PF cakes are the quickest way to test if an isolate has the traits you want.
But what's the quickest way to go from an isolate on agar to PF cakes?
I realize you could take a slice of the isolate and make a grain LC to knock up some PF jars, but is that how most of you do it?
Is there another way that's faster that I'm missing?

But how do you go from the isolate on agar to those? Is knocking up a grain jar with an agar wedge the only way?

you -could- simply exclude the vermiculite layer in your pf jars, giving additional space to stick your wedge in.
making lc/putting the wedge in grains is more ideal, i think.

Well, yeah, I mean "quick" relatively speaking.
I know everything in this takes patience. I just thought there might be another method to go straight to cakes or something, but I wasn't seeing it.

I do agar->lc or agar->grain->grain lc.
The step from agar to cake is the slow step of the process, the bottleneck.

You don't go quickly cuz there's no need to test agar. You're testing the LC. Can't test it till it's been made.
That's why when I receive a syringe the first thing I do is inoculate LC's with it and worry about isolation or cloning later.

That's pretty much what I thought, but I was hoping I was missing something here.
Thanks.

The fastest method of testing isolates is to make brf cakes without the verm layer, using a tyvek filter instead. Place a wedge of each strain in each brf cake. Don't waste time making LC(ever) or going to grains if the species you're testing will grow on cakes. RR
