P. Subaeruginosa cardboard tek project

Hello to everyone!

This is my first attempt at a cardboard tek with P. Subaeruginosa collected from a wild specimen.

I roughly followed the steps in ATWAR's tek (http://www.shroomery.org/8512/Cardboard-Cloning-Tek)  so thank you for the information you provided, I have supplied some further isolation for external contamination to improve my results and it seems to be working quite well.

I have take some pictures of my basic setup so you can see the steps I have followed to get where I am so far.

Project equipment list:

6 sealable plastic jars (small)
1 Plastic storage container
1 Sandwich bag
Cardboard (from a removals box)
Tap Water (ph 6.8)
Wild collected P. Sub mycelium, pins and mushrooms

I have done absolutely minimal sterilisation, other than washing the jars, containers and myself with dishwashing detergent.

I collected the mycelium from a small spot I found last year for research purposes only. It was gathered from 8 different sutes in a woodchip bed at a public garden facility. There were minimal mushrooms around at the time of collection, but the undergrowth was damp.

Method:

Soaked cardboard in normal tap water (which contains small amount of chlorine, ph 6.8 tested), and tore into 8cm x 8cm pieces, and seperated flat side of cardboard from corrugated, placed colonised woodchips containing mycelium (and some additional ground cover from the same site) onto the corrugated side and placed into moistened jar, adding about 5 layers to each jar and then sealing with lid. I placed by my aquarium to keep at optimal temp for mycelium production.

I checked all 8 seperate containers on the third day of project and there was ALREADY visible mycelium growth on the cardboard in about 5 of them. The other were just a little slow due to dryness of collected samples.

As you will see below the small jars tend to block the light a little better than the bigger container, but unusually the clearer container has the more prolific mycelium growth. The project in the sandwich bag dried up and failed to produce any noticeable mycelium so I would not recommend using any type of sealable bag for this tek as the seal is just not reliable enough for moisture retention.

I've not had to add any water after 7 days to the container, they seem to be retaining the moisture well, especially the larger container.

Also, I plan on transferring this mycelium to agar culture, once isolated.. I will then begin further projects on fruiting and cloning P. Sub and other local shrooms.

I dont have fruits as yet, the woodlovers are usually a challenge indoors.. its about to go out of season here, hence the timing of my mission!

I also have some questions for those who have experience in growing Subs or similar woodlovers indoors..
Do you think it would be advantageous to add substrate from the area I collected the mycelium to add the neccessary bacteria for fruiting and fast colonisation?
What type of chips would be colonised quicker indoors, dried chips or fresh chips?
How should I layer my fruiting case for correct bacteria colonisation and temperature control?
What type of temp & lighting cycle would be most beneifical for fruiting indoors? I have a fridge that can be used specifically for this.. :smile:

I think the amount of substrate that pulled up with your samples should have enough traces to boost the mycelium along.

Fresh chips are always better than older dried ones IMO. It takes out the step of having to hydrate them as well.

I have never actually fruited a woodloving Psilocybe before so I am not a pro or anything, I hope I helped a bit.

What I would do is wait for the cardboard to colonize and maybe put a thin casing of verm or peat moss and fruit maybe 1 or 2 like that.

I would also take a couple and do isolations on agar as you said, then do LC's and inoculate some bags/jars of grain, or do some sawdust/chips.

Thanks for the advice man! How do you suggest I do the isolation from cardboard to agar? I have petri's and a scalpel.

So today the growth is still going strong, especially at the bottom of the containers. I had to re-hydrate the cardboard, as it has dried from the constant opening and closing for gas release and fresh air. No contaminants are yet present but each container has a slightly different smell. Could be the collected substrate.

My agar kit should arrive this week so I will start 20 individual cultures in the hope that I can isolate the Sub mycelium for a few different inoculation projects.

This is going on a year now and it will be interesting to see how you go with this over winter.  Are you putting the containers outside?
