My Agar Work

I am trying to get a few good isolates and clones going on Agar. I am using MEA 12g to 300ml water + 4 ml h2o2
I have two samples, the one with the number 12 on it I would like some expert advice as to which section you would xfer.... The other is my cloning attempt uno  So what do you think, using the 12 as a reference, which section should i use, imagine the sample as a clock.

lmao your dishes are pretty much like mine cept i use crockery ... i thought i was the only ghetto-plate style agarist around here lol ... just make sure you get the utmost tips of those rhizos and nothing else so not to cloud the gene pool negligently ... will save you time in the end ...
ah shit man ur dishes aint ghetto thats something showing through ... thought those were designs on dinner plates lol

Id personally use the 1-3 just cause its faster growth and you still have time to sector for rhizo's, but 10-11 is nice growth

I'd make some transfers from all three sectors transferring each sector to it's own respective petri dish of agar and then see what kind of growth you get on each plate and make more transfers from the best looking sectors of each plate.
Can't wait till I get into working with agar!

Will listen to the experts then!!! MH I hope you can roll along with this thread and learn by watchin a noob roll. More info.... The peroxided agar does cause a stall out on the myc growth for sure. I do my work in a still box, and still ( lol no pun intended ) had contams. I did BOtH of thes transfers in very clean, yet open air...... So I figure from here on out until my FH is done it is MS -Milo - fruit - clone H2O2 agar -bulk
 very excited to finnaly be "getting" some of this!!!

what kind of contamination did you have? fungal or bacteriological?
peroxide will not solve any of your problems!

The growth in 10-12 looks strong.
Good luck!
P.S. For anyone who read the thread, I now have recovered all but two of my (what turned out to be) ten bacteria infected syringes. Each time I had to do at least two transfers for each strain. Now I have grain jars with wedges and am testing a wedge-based LC this evening. Agar rocks!

I've definitely done some reading on agar and strain isolation and have a pretty good idea about the whole process...but still,nothing beats hands on experience

That's why I am waiting until my lease is up and I get another place before I start agar work.At the moment the place I am in is too small and I have no where to place a flowhood.It's better for me to be patient and just wait for that moment in time as agar work in glove box.still air box would be very cramped,as well as g2g or a2g transfers.

The mexi cube would be the dirtiest plate... Just kidding haha, but very nice agar work. Its always nice to see that beautiful rhizomorphic stuff crawling across agar.
I feel pretty pro right now, I poured 25 dishes in a glove box, stored them in the fridge for 2 weeks, and have 25 sterile dishes now. After letting em sit out at room temp for a while. Yay me...
4 section dishes are so sexy and cheap, 4.70 a sleeve at cynmar.com just incase anyone needs them.

Yes I used Cyanmar too. I got some one and some three section dishes.
I also got some of the polypropylene dishes that a poster recommended.
I agree that agar works. I have had dishes out of the fridge for weeks and nothing has shown any contamination.
I may just go G2G with the grain jars that I have started with wedges. I have had some issues with LC...but I still love it.
